Зв’язок стимуляції антиоксидантного захисту рослин із ознаками нестабільності геному

На двох генотипах ромашки аптечної – сорту Перлина лісостепу та його мутанту досліджено зв’язок стимуляції нагромадження  низькомолекулярних антиоксидантів у фармацевтичній сировині рослин (суцвіттях) з ознаками радіаційно-індукованої нестабільності геному на стадії цвітіння рослин за умов рентгенівського передпосівного опромінення насіння при дозах 5–15 Грей. Дослідження перебудов первинної структури  ДНК за умов різних доз опромінення здійснено  шляхом проведення ПЛР з використанням восьми ISSR- та десяти RAPD–праймерів. Залежні від дози опромінення зміни спектрів ампліконів при проведенні ISSR – RAPD – ПЛР проаналізовано з використанням  індексу подібності Жаккарда. Встановлено,  що найбільші  перебудови первинної структури ДНК обох генотипів, що проявляється у зниженні подібності з контрольними спектрами ампліконів,  спостерігається при  дозах опромінення 5–10 Грей. Виявлено тенденцію до наближення цього показника до контрольного при дозі опромінення 15 Грей, що означає підвищення ефективності репаративних процесів. Показано зв’язок між поліморфізмом первинної структури ДНК за ISSR-RAPD-послідовностями у різних генотипів та характером її перебудови при  опроміненні. Співставлення цих результатів з немонотонними дозовими залежностями змін питомого вмісту флавоноїдів і фенолів, дозволяє зробити висновок про стимуляцію антиоксидантного захисту при дозах, що відповідають низький ефективності репараційних процесів і, відповідно, його зниженню  за умов ознак відновлення нормального стану генетичного матеріалу. Надано інтерпретацію виявленого феномену на основі відомого зв’язку ефектів геномної нестабільності з підвищенням рівня активних форм кисню і загальних принципів антиоксидантного захисту.  Обговорюється значення одержаних результатів в розробці наукового підгрунтя використання малих доз іонізуючого випромінення в біотехнологіях, зокрема в фармакології.
