Колокалізація білка BCR з клатрином, актином та кортактином свідчить про його можливу роль в регуляції розгалуження актину та клатрин-опосередкованому ендоцитозі

Філадельфійська хромосома є результатом взаємної транслокації між 9 і 22 хромосомами і є чітким маркером декількох типів мієлопроліферативних розладів. Така транслокація породжує різні типи злиття генів bcr та abl. Ці злиття відрізняються наявністю або відсутністю певних типів доменів BCR та молекулярною масою відповідних химерних білків. BCR-ABLp230 асоціюється з хронічним нейтрофільним лейкозом, BCR-ABLp210 – з хронічним мієлолейкозом. BCR-ABLp190 – з гострим лімфобластним лейкозом. Домен гомології плекстрину BCR присутній у p210 і відсутній у p190 типу злитого білка. У цій роботі ми показали, що білок BCR колокалізується з актином та кортактином на периферії клітин клітин K562. Цей білок також утворював скупчення колокалізації з клатрином та контактином і знаходився в точках розгалуження актину. Це було підтверджено мікроскопією STED із надвисокою роздільною здатністю та звичайною конфокальною мікроскопією живих клітин HEK 293T. Конфокальна мікроскопія в реальному часі також виявила відносно велику структуру в цитоплазмі, де спостерігалося динамічне сполучення BCR, клатрину та актину. Це сильно нагадує комплекс Гольджі, оскільки мережа транс-Гольджі є типовим місцем сортування та збирання пухирців, покритих клатрином. Отримані нами дані вказують на те, що BCR в тандемі з кортактином може відігравати важливу роль у динамічних перебудовах актину та мембрани, які впливають на опосередкований клатрином ендоцитоз та везикулярний транспорт Гольджі. Порушення його функції через аномальну активність тирозинкінази ABL може сприяти фенотипу раку.
