BARR Novel genetic variant of 30­bp deletion: a polymorphism of latent membrane protein 1 from vietnamese epstein virus­associated nasopharyngeal carcinoma biopsies

РЕЗЮМЕ. Назофарингіальна карцинома (НФК) – це найпоширеніший серед видів раку голови і шиї, які поширюються Азією. НФК тісно пов’язана з генною експресією віруса Епштейна­Барра (ВЕБ). Ген латентної мембрани білку 1 (LMP-1) і його варіант з делецією 30 п.н. мають значний вплив на багато шляхів патогенезу НФК, що призводять до онкогенезу. У В’єтнамі, країні з високим рівнем ендемії НФК, не проводилися дослідження варіанту з делецією 30 п.н. Таким чином, мета цього дослідження полягала у проведенні аналізу генетичних характеристик асоціації між делецією 30 п.н. у біопсії пухлин НФК і ВЕБ. Було отримано двадцять зразків біопсії НФК від місцевих пацієнтів, проаналізовано за допомогою ПЛР-секвенування і порівняно з прототипним варіантом делеції 30 п.н. B95-8. Внаслідок цього було зареєст­ровано поліморфізм делеції 30 п.н. у 55,00 % зразків. Поліморфізм делеції 30 п.н. було отримано від нуклеотиду 168,285 до нуклеотиду 168,256, що відрізнялися від прототипного B95-8 та інших послідовностей, про які повідомлялося раніше. Отже, ми це назвали новою делецією 30 п.н., виявленою у регіоні гену CDS LMP-1. Було зареєстровано три схеми заміни/делеції амінокислот, які спостерігалися у делеції 30 п.н./відсутності делеції 30 п.н. у C-цитоплазматичному кінці LMP-1. Новий варіант делеції 30 п.н.*, відмінний від попередньо повідомленої делеції 30 п.н., а також схема заміни/делеції амінокислот досліджували у зразках біопсії назофарингіальної карциноми у В’єтнамі.
