Effect of Co­treatment with static magnetic field and Cis­diamminedichloroplatinum (II) on apoptosis and cell cycle progression in HeLa cell line and Hu02

РЕЗЮМЕ. У цьому дослідженні нами було оцінено поєднаний вплив статичного магнітного поля і цис­диаміндихлорплатини (II) (CDDP) на загибель клітин, апоптоз і клітинний цикл у нормальних дермальних фібробластів (Hu02) та клітин HeLa. Клітини були піддані впливу CDDP у присутності 10 мТл статичного магнітного поля (SMF) впродовж 24 і 48 год. IC50 концентрації CDDP було визначено за опомогою МТТ­тесту. Цитотоксичний вплив такого одночасного застосування вивчали за допомогою проточного цитометричного аналізу для клітинного циклу і апоптозу. Отримані результати вказують на те, що одночасне застосування CDDP та статичного магнітного поля впродовж 48 год призвело до значного підвищення рівня зупинки прогресії клітинного циклу у фазі G2/M, некрозу і пізнього апоптозу у клітин HeLa порівняно з групою, яку обробили за допомогою CDDP. Натомість, таке поєднання призвело до зниження рівня зупинки у фазі G2/M, некрозу клітин у Hu02 порівняно з клітинами, обробленими препаратом. У присутності SMF чутливість ракових клітин до CDDP була підвищеною, хоча спостерігався деякий ефект захисту нормальних клітин від цитотоксичності препарату. Оскільки було виявлено очевидну відмінність між обробленими клітинами HeLa і Hu02 при поєднаному застосуванні SMF і CDDP на етапах клітинного циклу та апоптозу, можна висунути припущення, що спільне застосування SMF і CDDP може бути корисним способом зменшення побічної дії протиракового препарату на нормальні клітини та зниження стійкості ракових клітин до цього препарату.
