Морфологічні та молекулярно-цитогенетичні характеристики гігантських ооцитів людини

Питання доцільності використання ооцитів з морфологічним дизморфізмом у програмах допоміжних репродуктивних технологій є дискусійним. Метою роботи була оцінка морфологічних та молекулярно-цитогенетичних характеристик збільшених за розмірами ооцитів та прогнозування їх подальшого використання для запліднення. Із 774 ооцитів, вилучених у 80 пацієнток, 83 (11 %) – мали збільшений розмір. За даними морфометричного аналізу діаметр ооплазми становив (176,67 ± 1,76) мкм, з урахуванням ZP – (200,8 ± 4,24) мкм. На стадії метафаза ІІ знаходилися 47 (56,6 %) гіганських ооцитів, метафаза І – 26 (31,3 %), профаза – 10 (12,1 %). Вилучені гігант-ські ооцити характеризувалися дизморфізмом ендота екзоцитоплазматичних структур. На стадії метафаза ІІ знаходилися 47 клітин, з них 40 (85,1 %) ооцитів мали 2 полярних тіла, 3 (6,4 %) – 1 полярне тіло, а в 4-х (8,5 %) клітинах полярне тіло було фрагментоване. Методом поляризаційної мікроскопії у 39-ти (83 %) ооцитах візуалізували 2 веретена поділу, в 3-х (6,4 %) – 1, в 5-ти (10,6 %) ооцитах – не візуалізували, хоча полярні тіла були присутні. Після запліднення 47 ооцитів на стадії метафаза ІІ пронуклеуси було виявлено у 42-х (89,4 %) клітинах, з них в 9-ти (21,4 %) ооцитах візуалізували 2 пронуклеуси, в 27-х (63,3 %) – 3, в 6-х (15,3 %) – 4. Методом молекулярно-цитогенетичного аналізу було доведено, що ембріони, отримані після запліднення гігантських ооцитів, мають поліплоїдний набір хромосом. Таким чином, результати нашого дослідження доводять недоцільність використання збільшених за розмірами ооцитів для подальшого їх запліднення.
