Molecular characterization and tissue expression of common tobacco (Nicotiana tabacum) cadmium resistance protein 2 and metal tolerance protein 4 genes

РЕЗЮМЕ. ПЛР-ЗТ було використано з метою ампліфікації повних кодуючих послідовностей генів білку 2 стійкості до кадмію та білку 4 толерантності до металів у тканинах тютюну справжнього. Ген білку тютюну 2 стійкості до кадмію кодує білок з 192 амінокислот, який має високий рівень ідентичності з білком 2 стійкості до кадмію чотирьох видів рослин – Nicotiana tomentosiformis (99 %), тютюн лісовий (97 %), Capsicum annuum (90 %) та картопля (86 %). Ген білку тютюну 4 толерантності до металів кодує білок з 415 амінокислот, який має високий рівень ідентичності з білком 4 толерантності до металів шести видів рослин – Nicotiana tomentosiformis(90 %), тютюн лісовий (90 %), Lycopersicon esculentum (84 %), картопля (84 %), Lycopersicon pennellii (83 %) та Capsicum annuum (82 %). Передбачення трансмембранних спіралей продемонструвало, що білок 2 стійкості до кадмію та білок 4 толерантності до металів можуть бути двома трансмембранними білками. Філогенетичний аналіз показав, що і ген білку 2 стійкості до кадмію, і ген білку 4 толерантності до металів тютюну справжнього мають тісніший генетичний зв’язок з генами білку 2 стійкості до кадмію та білку 4 толерантності до металів тютюну лісового. Автоматизований аналіз продемонстрував, що ген білку 2 стійкості до кадмію є структурованим у 4 екзонах і 3 інтронах, а ген білку 4 стійкості до металів складається з 7 екзонів і 6 інтронів. Аналіз профілю експресії генів показав високий рівень експресії генів білку 2 стійкості до кадмію та білку 4 толерантності до металів тютюну справжнього у коренях.
