Isolation of antimicrobials from native plants of taif governorate

РЕЗЮМЕ. Дефенсини рослин – це підгрупа рослинних антимікробних пептидів з високим потенціалом для розвитку безпечних природних сільськогосподарських та фармацевтичних продуктів для біоконтролю як альтернативи наявним хімічним пестицидам, інсектицидам та антибіотикам. Полімеразно-ланцюгова реакція з використанням зворотної транскрипції (ПЛР-ЗТ) була використана у цьому дослідженні з метою проведення оцінки рівня експресії генів дефенсинів рослин у бутонах квітів 9 аборигенних рослин з Саудівської Аравії. Оскільки експресія дефенсинів активно проходить на рівні мРНК в Ochradenus baccatus (Ob), цю рослину було використано для виділення пептидів дефенсинів за допомогою іонообмінної хроматографії з наступною гель-фільтрацією за використання Sephadex G50. ДСН-ПААГ білків, очищених шляхом іонообмінної хроматографії, виявив декілька смужок у діапазоні 2–10 кДа. Ці білки були виділені шляхом гель-фільтрації на Sephadex G50. Виявилось, що отриманий 5 кДа пептид мав антимікробну дію. Він пригнічував грам-позитивні та грам-негативні бактерії, причому концентрація напівмаксимального інгібування (IC50) складала від 10 до 25 мкг/мл. Також цей пептид пригнічував від 25 до 60 % лінійного росту грибків при концентрації в 12,5 та 25 мкг/мл. Наші результати демонструють можливість виділення та очищення 5 кДа пептиду з Ob. Цей пептид може бути використаний для розробки антимікробних препаратів.
