Degradation of methanol catabolism enzymes of formaldehyde dehydrogenase and formate dehydrogenase in methylotrophic yeast Komagataella phaffii

З’ясування механізмів деградації цитозольних білків має велике фундаментальне та прикладне значення. Досліджено зміни в активності цитозольних ферментів катаболізму метанолу – формальдегіддегідрогенази (Fldh1) та форміатдегідрогенази (Fdh1) у контрольного штаму дикого типу GS200, штаму з делецією гена сенсора гексоз GSS1 та штаму з дефектом автофагії SMD1163 K. phaffii при короткотривалій та довготривалій індукції метанолом з додаванням або ж без додавання інгібітора протеасомної деградації MG132. Показано, що тривалість інкубації клітин на метанолі не має особливого впливу на інактивацію ферменту. Ефект протеасомного інгібітора MG132 був незначний. Катаболітна інактивація цитозольних і пероксисомних ферментів пошкоджена в gss1∆ мутанта, оскільки пошкоджене сигналювання глюкозою. Fldh1 та Fdh1 ймовірно деградують вакуолярним шляхом, незалежно від тривалості індукції метанолом.
