Most studies evidenced that Ras/Raf/MEK/ERK pathway promote proliferation and malignant transformation due to the stimulation of cell growth and division and apoptosis prevention. Signaling cascade PI3K/PDK/Akt is involved in the regulation of protein synthesis and cell energy supply. Besides, this signaling cascade inhibits apoptosis, promotes the survival of tumor cells and is activated in many types of cancer. However, hyperactivity of these cascades in tumor tissues can cause oncogene-induced senescence (OIS), growth arrest and apoptosis. This phenomenon was called «oncogene toxicity». The importance and mechanisms of oncogenic toxicity in thyroid and other types of cancers are discussed. Key words: oncogenes, malignant tumors, thyroid gland, MAPK, PI3K/Akt
