Apoptosis and processes of DNA fragmentation in native and cryopreserved human sperm cells at normo- and pathospermia

SUMMARY. In the treatment of infertility by methods of assisted reproductive technologies cryopreserved human sperm widely used. Carrying out a detailed study of the state of sperm cells after cryopreservation and subsequent cultivation will increase the effectiveness of assisted reproductive technology programs. The aim of study was investigated the effect of cryopreservation factors on the state of DNA and apoptosis in isolated fraction of human sperm at the normozoospermia and pathospermia. The DNA fragmentation analysis and evaluation processes of apoptosis, necrosis of spermatozoa were performed by methods of sperm chromatin dispersion and flow cytometry. In our study, cryopreservation did not affect DNA fragmentation when normozoospermia, but sperm cryopreservation at oligoastenoteratozoospermia significantly increased that index. The increase of apoptosis and necrosis processes of sperm after incubation at oligoastenoteratozoospermia gives reason for the choice of an alternative technique of oocyte fertilization – intra cytoplasmic sperm injection.
